اثر سالیسیلیک اسید بر صفات مورفو - فیزیولوژیک به لیمو (Lippia citriodora) در شرایط مزرعه ای و درون شیشه ای

چکیده

سالیسیلیک اسید یک ترکیب فنلى طبیعى و شبه­هورمون بوده و غلظت­های مختلف و مدت زمان تأثیرگذاری آن، کنش و واکنش­های متعددی را در گیاه سبب می­شود. در این راستا، دو آزمایش جداگانه به منظور بررسی اثر سالیسیلیک اسید بر صفات مورفو - فیزیولوژیک گیاه زینتی – دارویی به­لیمو در شرایط درون شیشه­ای و مزرعه­ای در شهرستان میانه انجام شد. در شرایط کشت بافت، پنج غلظت صفر، 5/2، 5، 10 و 20 میکرومولار سالیسیلیک اسید به محیط کشت موراشیگ و اسکوگ (MS) اضافه و اثر آنها بر شاخص­های رشدی به­لیمو مورد ارزیابی قرار گرفت. اثر سالیسیلیک اسید در شرایط کشت درون شیشه­ای اختلاف آماری معنی­داری در تمام صفات به جز در تعداد گره بلندترین ساقه، ارتفاع بوته و سیترال تولیدی نشان داد. همزمان با افزایش غلظت سالیسیلیک اسید، تعداد برگ­های کلروزه (زرد) ریزنمونه­های به­لیمو افزایش و درصد گیاهان سبز سالم و تعداد شاخه جانبی کاهش یافتند ولی در غلظت­های کم، طول بلندترین شاخه، تعداد برگ و شاخص کلروفیل برگ افزایش یافت. همچنین، نقش مثبت سالیسیلیک اسید در افزایش وزن تر شاخساره ریزنمونه­های کشت بافتی مشـاهده شد. در شـرایط مـزرعه­ای، اثر شش تیمار بـدون محلول­پاشی، محلول­پاشی با آب مقطر و سالسیسلیک اسید در غلظت­های 5/2، 5، 10 و 20 میکرومولار بر صفات مورفو-فیزیولوژیک به­لیمو مورد ارزیابی قرار گرفت. اثر سالیسیلیک اسید اختلاف آماری معنی­داری بین تیمارها در پهنای (عرض) برگ، تعداد گل­آذین، وزن تر گل­آذین، وزن تر برگ و طول بزرگ­ترین گل­آذین نشان داد. اثر منفی مصرف سالیسیلیک اسید در پهنای برگ، تعداد و وزن تر گل­آذین دیده شد و برعکس، وزن تر برگ و طول گل­آذین به­خصوص در غلظت کم سالیسیلیک اسید افزایش پیدا کرد. بنابراین، استفاده از غلظت­های کم سالیسیلیک اسید به خاطر داشتن اثر مثبت بر رشد اندام­های مصرفی به لیمو در شرایط مزرعه­ای و کشت بافتی قابل توصیه می­باشد.

Salicylic acid (SA) is a natural phenolic and a hormone like compound, and its different concentrations and treatment duration cause many actions and reactions in plants. Thus, two experiments were conducted to evaluate the effect of salicylic acid on morphological and physiological characteristics of lemon verbena as an ornamental and medicinal plant in in vitro and field conditions at Miyaneh, Iran. In tissue culture conditions, five concentrations of SA including 0, 2.5, 5, 10 and 20 μM were added to MS culture medium and their effects on lemon verbena growth indices were evaluated. Increasing SA concentrations caused increment in chlorotic leaf number but decrement in normal plantlet percentage and lateral shoots number in lemon verbena explants. However, in low concentrations of SA, longest shoot length, leaf number and chlorophyll content were increased. Although all concentrations of SA improved shoot fresh weight in lemon verbena explants. Under field condition, the effect of salicylic acid at 2.5, 5, 10 and 20 μM concentrations, non spray and spray with distilled water, were evaluated on morphological and physiological traits of lemon verbena. Salicylic acid affected significantly leaf width, inflorescence number, fresh inflorescence and leaf weights and the longest inflorescence length. The negative effect of SA application on leaf width and number and fresh weight of inflorescence was observed. But leaf fresh weight and inflorescence length improved especially in low SA concentrations. Therefore, application of low SA concentrations, because of their enhancing potential effect on vegetative growth of lemon verbena both in field and tissue culture conditions can be recommended.
